The only way to farm Agility Spheres is by defeating Fenrir at the Monster Arena. You can find two Agility Spheres in various chests across Spira, but those are only one-time treasures.
So Fenrir is the only way to farm them repeatedly.
And you can unlock Fenrir after capturing three of every Lupine-type monster on Spira. There are 7 different fiends of this type that you’ll need to catch, but remember that you’ll need x3 copies of each one.

Agility Sphere Uses
Agility Spheres are used to change an empty node on the Sphere Grid into an Agility +4 node.
You’ll then need Speed Spheres to activate any Agility nodes you create.
Other than creating Agility nodes, Agility Spheres can also be used in some of Rikku’s mixes. Here’s a few of the more interesting mixes you could try:
FreedomX (Three Stars + Agility Sphere): Reduces MP cost for all abilities to 0 for the party.
Megalixir (Al Bhed Potion + Agility Sphere): Restores party HP and MP to full.
Final Phoenix (Mega Phoenix + Agility Sphere): Revives all KO’d party members to full HP and MP.
Are These Spheres Worth Farming?
No, not really.
The only thing Agility dictates is how often a character gets their turn in battle. And while that is important, it has a hard decrease in effectiveness at 170 Agility – and that’s a number you can get to just by activating and powering up all the already-existing agility nodes on the grid.
Empty spaces are more important for Luck and Evasion boosts.
Even Rikku’s mixes that Agility Spheres are a part of can be created with other items that are much easier to obtain.
